Biogenesis of microRNAs (miRNAs) can be summarized in five steps (reviewed in Ketting 2011, Nowotny and Yang 2009, Kim et al. 2009, Chua et al. 2009, Hannon and He 2004):1. Studies suggest that approximately half of known microRNA reside in non-protein coding RNAs (intron and extron) or within the intron of protein coding genes (Rodriquez et al. 2004).

MicroRNA (miRNA) biogenesis requires AtDRB1 (double‐stranded RNA binding protein)/HYL1 (Hyponastic Leaves1) protein for processing and maturation of miRNA precursors. The AtDRB1/HYL1 protein associates with AtDCL1 (Dicer‐Like1) and accurately processes primary‐miRNAs (pri‐mRNAs) first to precursor‐miRNAs (pre‐miRNAs) and finally to mature miRNAs.
